Nepafenac is a novel ocular non-steroidal antipyretic, analgesic and anti-inflammatory drug (NSAIDs). Nepafenac is used to treat eye pain and inflammation caused by cataract surgery. Compared with traditional NSAIDs, the chemical structure of Nepafenac is favorable for the drug to rapidly penetrate the cornea and distribute at its target site. Function Of Nepafenac

Nepafenamine, when administered via the eye, rapidly passes through the cornea and is converted to amifenic acid (an NSAID) by hydrolases in the eye tissue; Amifenic acid plays an anti-inflammatory and analgesic role by inhibiting prostaglandin H synthetase (cyclooxygenase) and blocking the synthesis of prostaglandin. As is well known, prostaglandins are mediators that cause inflammation in the eye, leading to breakdown of the blood-aqueous barrier, vasodilation, increased vascular permeability and leukocyte chemotactic, etc. In addition, prostaglandins can also contract the iris sphincter through non-cholinergic mechanisms, leading to the constriction of the eyes during and after ophthalmic surgery. When administered via the eye, NSAIDs inhibit the synthesis of prostaglandins in the iris, ciliary body, and conjunctiva, thereby preventing eye inflammation and reducing associated pain.
