What is Delafloxacin?

Chemical Classification

Fluoroquinolone antibiotic (4th generation modified), chemical name: Delafloxacin.

Structural features: Amphoteric ionic structure (coexistence of acidic/basic groups), enhanced tissue penetration and target binding.

Developed in response to the escalation of resistance to traditional quinolones, it enhances the therapeutic efficacy against gram-positive bacteria (e.g. MRSA) and complex infections.

Antimicrobial Spectrum Properties

Broad spectrum coverage:

Gram-positive bacteria: Staphylococcus aureus (including MRSA), Streptococcus pneumoniae.

Gram-negative bacteria: Escherichia coli, Pseudomonas aeruginosa, Haemophilus influenzae.

Atypical pathogens: Mycoplasma pneumoniae, Chlamydia.

Anti-drug resistance advantage: maintain high susceptibility to multi-drug resistant bacteria (e.g. methicillin-resistant strains).

Mechanism of action

Dual target inhibition:

Inhibits bacterial DNA rotamase (topoisomerase II) and topoisomerase IV, blocking DNA replication and repair.

Stronger target binding to resistant bacteria than other quinolones.

Pharmacodynamic (PD) characteristics

Concentration-dependent bactericidal activity, high dose short course strategy recommended to maximise efficacy and reduce risk of resistance.
