Depression, also known as depressive disorder, is a major type of mood disorder with significant and persistent low mood as the main clinical feature.

Paroxetine is a phenylpiperidine derivative with the molecular formula C19H20FNO3 and its specific structure is shown in Figure 1. It is the SSRIs that selectively inhibit 5-HT transporters, block the reuptake of 5-HT by the presynaptic membrane, prolong and augment the effects of 5-HT, and produce antidepressant effects. At typical doses, it has no significant effect on other mediators, except for weak inhibition of NA and DA reuptake. It is completely absorbed after oral administration, with a bioavailability of 50%, and food and pharmaceuticals do not affect absorption. It is metabolized primarily in the liver to produce uridate compounds, some of which are mediated by CYP2D6. Finally, it is excreted by the kidneys and a small amount is excreted in the feces by biliary secretion. The effective plasma concentration reference range for paroxetine is 20.0 to 65.0 ng/mL, with a laboratory warning concentration of 120 ng/mL. Common side effects include mild dry mouth, nausea, loss of appetite, constipation, headache, tremors, fatigue, insomnia, and sexual dysfunction. Sometimes neurogenic edema, urticaria, and orthostatic hypotension. Rare reports of extrapyramidal responses.
