Overview:Ritonavir is an HIV protease inhibitor developed by Abbott of the United States and is used clinically for the treatment of AIDS.

Function

Ritonavir is an HIV protease inhibitor that interferes with the reproductive cycle of HIV. Although it was initially developed as an independent antiviral agent, it has been shown to possess advantageous properties in combination regimens with low-dose ritonavir and other protease inhibitors. It is now more commonly used as a booster of other protease inhibitors and is available in both liquid formulation and as capsules.

The main usage Ritonavir is to treat patients with human immunodeficiency virus (HIV), specifically Type 1, the more dangerous and prevalent type of HIV. Usually, Ritonavir and Lopinavir are combined in a tablet or in a liquid solution to make the treatment more effective. Basically, lopinavir and ritonavir act as an antivirus, preventing and reducing the production of HIV virus in the body. Patients should be informed that lopinavir and ritonavir are not used as a cure for HIV; rather, they are only used to treat and manage the condition. The combined drugs also do not prevent the virus from being transmitted from one person to another.

Compound(I)wasdissolvedindichloromethane,andthedichloromethanesolutionofcompound(II)andtriethylaminewasaddedundertheprotectionofnitrogenat-15℃,andstirredfor2h.Theresultingmixturecontainedcompound(III)anduseddirectlyNextstepreaction.Slowlyaddtriethylaminetotheabovereactionsolution,andaddtheresultingmixturetothedichloromethanesolutionoftriphosgeneat-15to13°C,andreactfor1.5hours.Thedichloromethanesolutionofcompound(IV)wasslowlyadded,andthereactionwascarriedoutfor1hour.Waterwasadded,theorganiclayerwasseparated,washedwith10%citricacidaqueoussolution,andconcentrated.Theresiduewaspurifiedwithsilicageltoobtaincompound(V),andtheyieldbasedoncompound(I)was38%.Compound(V)isdissolvedinaceticacidandhydrogenatedunderpalladiumcatalysis.FilteChemicalbookrandconcentrateunderreducedpressure.TheresidueisdissolvedinwaterandadjustedtoPh=8withsodiumhydroxide.Itwasextractedwithdichloromethane,andtheextractwaswashedwithwaterandconcentratedunderreducedpressuretoobtaincompound(VI),whichwasdirectlyusedinthenextreaction.Thecompound(VII)wasdissolvedinethylacetate,sodiumbicarbonateaqueoussolutionwasadded,andthelayerswereseparatedafterthereaction.Theseparatedorganiclayerwasaddedtotheethylacetatesolutionofcompound(VI),andreactedat60°Cfor12hours.Concentrate,addammoniaandstirfor1h.Separatetheorganiclayerandwashwith10%potassiumcarbonateandsodiumchlorideaqueoussolutions.Itwasconcentratedunderreducedpressure,andtheresiduewaspurifiedwithsilicageltoobtaindritonavir.Theyieldbasedoncompound(V)was27%.
