Olaparib Usage

Olaparib plays an important role in DNA damage repair and cell apoptosis. Therefore, Olaparib specifically acts on the DNA damage repair mechanism of targeted cells and can be used for the maintenance treatment of patients with recurrent severe ovarian cancer who have breast cancer susceptibility gene (BRCA) mutations and are sensitive to platinum drugs.

Olaparib is a potent inhibitor of poly (ADP-ribose) polymerase (PARP). Olaparib has been shown to induce significant killing of ATM-deficient lymphoid tumor cells in vitro and in vivo. Recent studies have shown that Olaparib can enhance the radiosensitivity of lung cancer tumor xenografts, making it a potential candidate for use in combination with radiation therapy.

Olaparib effect

Olaparib is effective against Brca1;p53 breast tumors (50 mg/kg/day intraperitoneal injection), but not against HR-deficient Ecad;p53 breast tumors. Olaparib has no dose-limiting toxicity in tumor-bearing mice. Olaparib has been used to treat BRCA-mutated tumors, such as ovarian, breast, and prostate cancers. In addition, olaparib selectively inhibits ATM-deficient tumor cells, suggesting that olaparib may be a potential agent for the treatment of lymphoid tumors with ATM mutations.
